## Guide

### 1. Acetaminophen 500 mg: batch release

A GMP quality\-control laboratory releasing a batch of Acetaminophen Tablets USP, 500 mg\. Samples flow from receipt and LIMS registration through physical testing to a robotic preparation cell and the HPLC bay, with wet chemistry, stability and QA review behind\. Tap any instrument to frame it; drag elsewhere to turn the lab\.

Zones follow a one\-way flow with weighing and preparation beside chromatography and solvent storage next to the HPLC systems; microbiology is segregated with its own airflow\.

### 2. Where the assay sits in QC

Release testing runs in parallel: identification \(HPLC retention time, TLC or FTIR\), dissolution per USP <711>, uniformity of dosage units <905>, related substances and the assay\. Raw\-material, in\-process and stability testing bracket it\. Only when every result meets specification does QA sign the certificate of analysis\.

Monograph dissolution: Apparatus 2 at 50 rpm, 900 mL pH 5\.8 phosphate buffer, 30 min, Q = 80 %\. Titration remains the assay route for many drug substances; this tablet assay is HPLC\.

### 3. Sample receipt and LIMS

The batch sample is logged in LIMS against batch number, product and test list, labelled with a unique barcode and stored under its condition\. The tote then moves to the preparation cell, where the scheduler reads the barcode and pulls the assay method, specification and worklist from LIMS\.

Instrument results are later imported against the same sample ID, so no result is transcribed by hand\.

### 4. Robotic prep: weigh and extract

The homogeniser powders at least 20 tablets\. The gripper takes a 200 mL volumetric flask to the balance, where powder equivalent to about 100 mg acetaminophen is weighed, then to the dispenser for about 100 mL of mobile phase\. The flask is shaken for 10 min and sonicated for about 5 min\.

Every weight, volume and timestamp is written by the workstation software to the electronic record\. Automated tablet workstations have shown 99\.4–101\.1 % recovery with RSD below 0\.5 % \(PMC2548120\)\.

### 5. Dilute, filter, vial

The extract is diluted to volume, then 5\.0 mL is transferred to a 250 mL flask and diluted again: a dilution factor of 10,000 to about 0\.01 mg/mL\. It is filtered through 0\.5 µm or finer, the first 10 mL discarded, and the capped, barcoded vial loaded to the autosampler tray\.

Discarding the first filtrate saturates any adsorption sites on the membrane so the vial concentration matches the flask\.

### 6. The HPLC stack

Isocratic reversed\-phase conditions: methanol:water 1:3 at 1\.5 mL/min through an L1 \(C18\) column, 3\.9 mm × 30 cm, 10 µL injected and UV detection at 243 nm\. The pump draws from the solvent tray, the autosampler injects into the flow, and the column, outlined in black, separates acetaminophen before the detector\.

Conditions from the USP acetaminophen monographs; current revisions allow modernised columns within USP <621> adjustments\. Footprint of an integrated system is about 50 × 63 × 74 cm\.

### 7. System suitability

Before any sample counts, replicate injections of USP Acetaminophen RS must show tailing factor NMT 2\.0, column efficiency NLT 1000 theoretical plates and RSD NMT 2\.0 %\. Age the column with the slider and watch the peak tail, plates fall and the run fail\. A failed suitability invalidates the run; samples are not reinjected until it passes\.

Acceptance limits are the monograph's; the degradation curve against column usage is illustrative\.

### 8. Sample injection and result

Bracketed by standards, each sample preparation is injected and integrated\. Content in the weighed portion is 10,000 × C × rU/rS mg, with C the standard concentration in mg/mL and rU/rS the sample\-to\-standard peak response, reported as percent of label claim\. Specification: 90\.0–110\.0 %\. Inject preparations and watch the spread\.

A result outside the limits is out of specification: it is not retested away but opens a documented OOS investigation in LIMS\.

### 9. Records, review and release

The chromatography data system, such as Waters Empower in a validated 21 CFR Part 11 configuration, keeps the raw data and a secure, time\-stamped audit trail of user, run, integration and reprocessing\. A second analyst reviews data with its audit trail, LIMS assembles the CoA, and QA releases the batch\. Toggle a manual reintegration to see it recorded\.

FDA Data Integrity Q&A \(2018\): audit trails are reviewed with the record, 21 CFR 211\.68 requires exact backups and access control, 211\.194\(a\)\(8\) a second\-person review\. ALCOA\+ throughout\.
